744 Evaluation of lipid ordering in the stratum corneum and lipid fluidity in the stratum granulosum using two-photon microscopy

Topical application of polyols such as xylitol and fructose accelerated epidermal water-impermeable barrier recovery. The polyoxyethylene/polyoxypropylene dimethyl ether (EPDME) random copolymer accelerates recovery the well. Previous physicochemical studies have demonstrated that these polymer interact with phospholipid membrane. To examine interaction between polyols/polymer lipids in epidermis, we stained epidermal-equivalent model Laurdan, a fluorescent dye to detect physical properties membrane environment, carried out two-photon microscopic observation. First, examined changes lipid fluidity stratum granulosum after solutions. solutions increased uppermost part exocytosis lamellar bodies intercellular domain corneum granulosum. Then, observed ordering EPDME. EPDME solution improved at upper prevented penetration water-soluble dye. In summary, increase increases corneum. Application xylitol, fructose, EPDME, which leads improvement function, might improve homeostasis by different mechanisms.
